Buflomedilhydroc Hloride Injection

[Pharmacology and Toxicology] Buflomedil Hydrochloride is a α-adrenergic receptor inhibitor, with weak nonspecific calcium ion antagonism. It can improve the blood flow of microcirculation in brain and limbs by inhibiting precapillary sphincter spasm. It also has a function of inhibiting platelet aggregation and improving red cell deformability. It has been reported in documents that, the non-toxic dose for rat by intravenous injection is 12mg/kg/day in a study on chronic toxicity. In addition, it has no mutagenesis and teratogenic effect.

[Pharmacokinetics] After intravenous injection, Buflomedil Hydrochloride is distributed widely in body fluid and tissues, metabolized and removed via liver with a half lifetime of about 3h. Buflomedil Hydrochloride is discharged via urine mainly in the form of original drug and metabolites with aromatic ring demethylated. The serum protein binding rate is related to the blood drug concentration. It is 81% and 25% when the blood drug concentration is 0.5mg/L and 5mg/L respectively

[Indications]

1. Peripheral vascular diseases: intermittent limping, Raynaud's syndrome, Burger syndrome and vascular spasm.

2. Symptoms caused by chronic insufficient blood supply to cerebral vessels: dizzy, tinnitus, intelligence impairment, memory impairment, impaired concentration and unorientation ,etc.

[Adverse reactions] gastro-intestinal upset (heartburn, stomachache, nausea), headache, dizziness, drowsiness, insomnia, scorching and tingling sensation, erubescence or pruritus. Palpitation, atrial fibrillation, elevated serum creatinine level, increased urine volume, change in menstrual blood volume, hypertension, nasal bleeding and psoriasis are uncommonly reported.

[Contraindications] Use of the drug is contraindicated in patients with hypersensitivity to it, in those with acute myocardial infarction, stenocardia, hyperthyrosis, paroxysmal tachycardia, cerebral hemorrhage, and in those with hemorrhagic tendency or with massive blood loss recently.
